Is there difference between point to point vs. other wiring methods? Is there really a sound difference?

No sound difference, one looks pretty, the others don't (unless you prefer PCBs, Vero or Perf).

Yeah, no sound difference. Except possibly some extreme cases. I believe proper PCB design can cut down on noise because of ground planes and such, but I'd be surprised if you heard a difference.
IMO, point to point makes it easy to visualize the circuit. And it certainly makes mods a bit easier. And, honestly, I don't think one method is harder than the other for small circuits. Once it gets more complex than a boost of Fuzz Face or something, though, PCBs are way faster.

I like making point to point stuff. Looks pretty, and like McSpunckle said, really easy to mod. Sometimes I make the base circuit and then just start adding parts to see how it sounds!

Anti-PCB tube amp fanatics get in tizzies about stray capacitance from parallel traces causing problems. Can also happen with bad point-to-point wire dress. The advantage to PCB there is once you get those bits of layout right, that part isn't going to create an unexpected problem.
Other than that, it's all preference, serviceability, and mod-ability if you're so inclined. (I can't deal with SMD myself -- too dang small for my fingers to deal with.)

Most true P to P looks like a mess unless the builder is anal.
Down side with all the components just hanging in the air a loose battery can wreak havoc. One of the joys of vintage stuff LOL.
Also unless you insolate the leads, they can come in contact with each other when vibrating.
here is an eyelet/P to P combo from the '60s
and a very neat eyelet layout.

PTP is real fun unless its for like a muff.

I prefer using turret board, though it has it's limitations/tribulations once you start building things bigger than fuzz face sized circuits...delays for instance...I'll have to get back to you on how that turns out.
